What is the difference between in and by?

I have these two sentences:
Austrian stamps was printed in a second step
Austrian stamps was printed by a second step
Which one is correct?
Can you tell me the difference between them?
I was stuck in finding and using these words correctly. Hope y'all help me. Thank you so much!

First of all: WERE printed (not WAS printed) - Austrian stamps is plural

Probably, if I understand your meaning:

Austrian stamps were printed in a second step

Please supply more context for a proper answer.
