It has been a long time since I didn't stay up this late at night.
Does this sentence make sense?
Could since introduce a negative sentence? If so, could you please give me some examples?

Unless you're talking in your sleep the sentence doesn't make sense - and even then it wouldn't really.

It does make sense if you drop "didn't" and use the past tense for "stay", however try using a more active voice when writing.

"I haven't stayed awake this late in months [days or years, whatever time period you're trying to convey]." or "I'm usually in bed by now."

i think this is correct,
'cause as I remember there was a sentence in our textbook "it has been a long time since we met last year"
i'm chinese :D
