Answer 1: David has two younger brothers, hasn't he?
Answer 2: David has two younger brothers, doesn't he?
Question 2:
My cousin has a wound on her knee.
Answer 1: My cousin has a wound on her knee, hasn't he?
Answer 2: My cousin has a wound on her knee, doesn't he?

Both question-tags are possible, but the first is generally restricted to BrE usage.
